Orthotopic Human Prostate Cancer Model in Athymic Nude Rat

• Human Prostate cancer (LnCaP) cell line
• Proprietary Immunomodulation of BRM ru/ru nude rat (SuperNude)
• Implanted into prostate of SuperNude/BRM rat strain (Athymic and lyp insert)
• Readouts: thru 8 weeks, Body Weight, Tumor Size at Necropsy and Histopathology relative to time post treatment

Protocol:

• Athymic male nu rats (90+ days)
• Group Size n=6
• Pre-treatment
  • Proprietary Treatment (Day -6, -4, and -2)
  • Survival Surgery (Day 1): inject 10⁶ LnCaP cells, left ventral prostate lobe
• Treatment Groups (2x weekly, Days 14-28)
  • Doxorubicin 5mg/kg, IV
  • Suramin 10mg/kg, IV
  • Doxorubicin + Suramin, IV
  • vehicle
• Analyses
  • Clinical: body weight and serum PSA, weekly
  • Terminal: Tumor Weight and Histopathology

Heterotopic Human Breast Ductal Carcinoma Model in Athymic Nude Mouse Overview

• Human Breast Cell Line BT-474
• Cultured cells injected into subcutaneous compartment of nude (athymic) mice
• Mice must be pretreated with 0.72mg/day 17 beta-estradiol implant
• Readouts: Tumor size by caliper
• Histopathology of major organ system

• Pre-treatment:
  • Estrogen Implant (Day -1, 0.72mg pellet/mouse)
  • BT-474 cells inoculated (Day 1, 10⁷ cells, SC)

  • Tumor Size (ranging from 0.2-0.3 cm³
  • Body Weight
• Treatment (Day 14-49):
  • Tumor Size (ranging from 0.2-0.3 cm³
